Ítem Acceso Abierto Comparación de dos crioprotectores en la congelación de espermatozoides de Jambato de las Tres Cruces del Azuay(Universidad Católica de Cuenca., 2024) Alvarado Lucero, Marjorie Janina; Argudo Garzón , Daniel Ernesto; 0704642818Cryopreservation is one of the most effective techniques for the long-term storage of genetic samples (gametes), in which cryoprotectants can be used to prevent the excessive accumulation of electrolytes and other substances during the freezing process and the creation of ice crystals that break the membrane structure. The objective of this study was to analyze two cryoprotectants, Dimethyl sulfoxide (DMSO) and Dimethylacetamide (DMA), at 10% in the freezing of spermatozoa from the species Atelopus Nanay of Azuay. The human chorionic gonadotropin hormone (hCG) was administered for sperm production in four individuals. The non-lethally collected spermatozoa were cryopreserved and unfrozen for subsequent evaluation. The use of cryoprotectants (10% DMSO and 10% DMA) showed no statistical difference in morphology. The 10% DMSO treatment showed 25.00 ± 50.00% in plasma membrane integrity and 37.50 ± 47.87% in mitochondrial functionality. Therefore, using both cryoprotectants at a concentration of 10% did not favor the preservation of the kinetic and qualitative characteristics. Keywords: Cryopreservation; Amphibiams; Sperm; DMSO; DMA.
